Simple Mug Test This is a simple reliable technique which uses a dry medium to enable the rapid confirmed enumeration of both total coliforms and E.coli.
The media turns blue-green in the presence of coliform bacteria (as illustrated in the above photo). E. coli growth is confirmed by blue fluorescence under UV light or using a Kovacs reagent.
For Coliform Confirmation: a) CLEAR/YELLOW = NEGATIVE for Coliforms
For E.Coli Confirmation: Shine a UV light (approx 365nm) from the bottom of the sample while avoiding looking directly at the light. a) NO FLUORESCENCE = NEGATIVE for E.Coli
If further confirmation of E.Coli is desired add 1ml of Kovac’s Indole reagent (sold separately) to the sample , recap the bottle and wait 30 seconds. A RED ring will form if E.Coli is present.
If results are positive add bleach to the sample before pouring down the toilet. |
